A team from São Paulo State University has found the exercise hormone named irisin to have the ability to fight COVID-19. The therapeutic effect of irisin is currently being explored to help save more lives. On studying adipose cell gene regulation, the researchers found a connection between irisin modulated genes and the replication of the coronavirus SARS-CoV-2 in human cells. The transcriptome was used to study the non-infected adipose cells. The new findings published in Molecular and Cellular Endocrinology support the actions of thyroid hormones and irisin present in adipocytes. The effects of irisin on the gene expression encouraged the researchers to test it on the replication of SARS-CoV-2.

The hormone was found to reduce the expression of the genes HAT1, TLR3, HDAC2, SIRT1, KDM5B, FURIN, RAB1A, and ADAM10 that stimulate ACE-2, a gene important for the virus to bind and invade the human cells. Furthermore, it was observed that irisin triggered a threefold increase in transcription of the gene TRIB3. The expression of TRIB3 is important but it is found to diminish with age and could be related to the increase in replication of SARS-CoV-2. This may be the major reason behind the elderly population easily falling prey to this pandemic.

In addition, the adipose tissue is found to be the repository for the virus and thereby explaining the risks in obese individuals. Furthermore, the irisin levels are lower whereas ACE-2 levels are higher in obese people. Continuous exercising is known to produce irisin that turns adipose tissue brown and this brown fat break down help maintain body temperature and create heat. The irisin can thus be used to treat metabolic disorders like obesity and also to modulate the macrophage activities. The researchers are working on converting their bioinformatics investigation into a 3D adipose cell model.
